2018 Annual Report – Letter from DirectorLetter from the NIEHS and NTP DirectorIn fiscal year (FY) 2018, NTP continued to advance toxicologyand inform public health policy by providing information todecision makers and the public about substances in ourenvironment. Numerous studies were published on substancesof public health concern, such as flame retardants, cell phoneradiofrequency radiation, and widely used industrial chemicals.Among these is the final report from the CLARITY-BPA corestudy and data sets from CLARITY-BPA grantee studies toevaluate the range of potential health effects from BPA in rats.In addition, NTP engaged in several activities to advancealternatives to animal testing. Particularly noteworthy waspublication of the Strategic Roadmap for Establishing NewApproaches to Evaluate the Safety of Chemicals and MedicalProducts in the United States, a consensus perspectivedeveloped with input from many groups. The “Tox21” consortiummet to discuss the new Toxicology in the 21st Century (Tox21)strategic plan that will continue to refine prediction of chemicaltoxicity to humans.NTP continued its systemic review literature activities to identifypotential hazards for human health. In partnership with the NIHCountermeasures Against Chemical Threats (CounterACT)Program, NTP conducted a systematic review to assess longterm neurological effects following acute exposure to theorganophosphorus nerve agent sarin. Antimony trioxide andHelicobacter pylori were evaluated for possible listing in thecongressionally mandated Report on Carcinogens.Dr. Birnbaum has served as the Director ofthe National Institute of EnvironmentalHealth Sciences (NIEHS) and the NationalToxicology Program (NTP) since 2009.(Photo courtesy of Steve McCaw.)I invite you to read this report to learn about our work and what weaccomplished in FY 2018 toward advancing toxicology andsafeguarding public health by generating and communicatingtrusted scientific information.Linda S. toxicology data would potentially be required. For example, as noted in the toxicology data requirements table, swimming pool, wood preservative, metalworking fluid, and aquatic areas uses require toxicology data in addition to the core toxicology data. Again, the use pattern and specific use site(s) should be examined to determine the data .

toxicology and carcinogenesis studies were conducted in F344/N rats by administering hexachloro- ethane (approximately 99%pure) in corn oil by gavage to groups of males and females for 16 days, 13 weeks, or 2 years. Genetic toxicology studies were conducted in Salmonella typhimurium and in Chi- nese hamster ovary (CHO) cells.

4 Toxicology Toxicology Terminology Toxicology is the study of the adverse effects of chemicals or physical agents on living organisms. A toxicologist is a scientist that determines the harmful effects of agents and the cellular, biochemical, and molecular mechanisms responsible for the effects. Toxicant, toxin, and poison are often used interchangeably in the literature; however,

The classic connotation of toxicology was "the science of poisons." Since that time, the science has expanded to encompass several disciplines. Toxicology is the study of the interaction between chemical agents and biological systems. While the subject of toxicology is quite complex, it is necessary to understand the basic concepts in order to make
