Pyrogen removalThe most common pyrogens are endotoxins,i.e., lipopolysaccharides (LPS) from the walls ofGramnegative bacteria. The LPS have two major parts:a hydrophilic polysaccharide chain with antigenicregions and a hydrophobic lipid group. As thepolysaccharide chain is variable in length, the LPSmolecular weight ranges from 3,000 to 25,000 Dalton.In ultrapure water, the LPS sub-units aggregate toform higher molecular weight structures that can beremoved by ultrafiltration membranes with cut offbelow 20,000 Dalton.Pyrogens are known to affect cell culture andbiochemistry experiments in numerous ways. Ithas been demonstrated that their interaction withcell membranes causes morphological changesand damage, as well as the secretion of specificsubstances such as tumor necrosis factor, cytokines orenzymes. Pyrogens also affect the cell division process(enhancing or reducing it) depending on the nature ofthe cell line. The presence of pyrogens also may affectanalytical techniques such as electrophoresis.For these reasons, it is good laboratory practice toremove pyrogens from all solutions used in cell cultureand other biochemical applications. Experimentsperformed in EMD Millipore R&D laboratorieshave demonstrated that the BioPak ultrafiltrationcartridge can be used for at least 90 days to treatMilli-Q ultrapure water and obtain product waterwith a pyrogen level below 0.001 EU/mL.Nuclease removalChallenge tests performed in EMD Millipore R&Dlaboratories have demonstrated that the BioPak cartridge allows easy production of ultrapure waterthat is both RNase-free ( 0.01 ng/mL) and DNasefree ( 4 pg/μL).Previous experiments also have shown thatultrafiltration with a properly validated device is justas efficient as diethylpyrocarbonate (DEPC) for RNaseremoval from ultrapure water without the negativeaspects of DEPC treatment: lengthy treatment timeand contamination of the treated water by CO2 andethanol.2

COMPARISON OF LC-PAK AND SOME LC-MS BOTTLED WATERIn order to verify that fresh ultrapure water produced by a Milli-Q system and an LC-Pak cartridge matchedor exceeded the quality of commercially available bottled water for LC-MS applications, different 60 mL watersamples were pre-concentrated on a C18 reverse phase chromatography column, then eluted and an lyzed byLC-MS (PDA, single quadrupole). Today, endocrine disrupters are increasinglypresent in the environment. Althoughendocrine disrupters may be only somewhatdangerous for adults, mounting evidencesuggests that they can significantly disturbfetal and infant development. As researchersincreasingly focus on the effects of thesechemicals, there is a need for endocrinedisrupter free water for use in experimentaltests. While conventional water purificationtechniques remove the bulk of organiccompounds from ultrapure water, some traceorganics still may remain.These man-made chemicals can be foundin many products widely used in industrialsocieties: Bisphenol A is a plasticizer used in reusablewater bottles, laptop computer housings,dental sealants and the resins in some foodcan linings. Phthalates are found in a wide varietyof products, including vinyl flooring, foodpackaging, blood-storage bags, intravenousmedical tubing, as well as many health andbeauty products such as detergents, soaps,shampoos, deodorants, fragrances, hair sprayand nail polish. Youare now ready to deliver at least 300 l of VOC-freeultrapure water.VOC-Pak POLISHER SPECIFICATIONSThe VOC-Pak cartridge is designed to producewaterin which the VOC concentration is below theanalytical quantification limit (**) for the VOCs listedin Table 1, provided that the VOC concentration infeed water does not exceed 1 ppb (1 μg/l), and theTotal Oxidizable Carbon (TOC) of the ultrapure waterentering the VOC-Pak cartridge is below 5 ppb.In order to warrant this water quality, theVOC-Pak cartridge was qualified by a challenge test:the VOC-Pak cartridge was fed by ultrapure watercontaining a 1 ppb (1 μg/l) concentration of the VOCto be eliminated.The concentration of the VOC was measured by GasChromatography (GC) downstream of the VOC-Pak cartridge after passage of 50, 100, 150, 200, 250,300 and 350 l in order to determine the breakthroughpoint of the different VOCs. The results are displayedin Table 1 for a processed volume of 350 l.Q-POD dispenser fitted with a VOC-Pak cartridge

Analysis of the VOCs in ultrapure water was performedaccording to EMD Millipore procedure 84214SO,using an Eclipse 4660 Purge-and-Trap sampleconcentrator from OI CorporationTM (College Station,Texas) and a GC-MS system (GC model 6890N and MSmodel 5975B) from Agilent Technologies , Inc., SantaClara, California.
